You can use Picrosirius Red Staining (PSR) + Polarized Light Microscopy that stains mature, cross-linked collagen (type I, II, III) red, while procollagen and newly synthesized collagen appear green or yellow under polarized light. You can expect Mature, thicker fibers to appear Bright red/orange Immature/thin fibers (including procollagen) to look Green/yellow This method is not dependent on immunohistochemistry distinguishing mature and immature collagen fibers.
