Have you tried using flow cytometry to track the cells, use a sorter to sort CAR T cells and assess their function using functional killing assays

Thanks for your reply. We checked the cells using Flow cytometry only and that’s how we came to the conclusion that cells are persisting for variable duration in different individuals.
What do you suggest we do further?
A suggestion could be to use flow cytometry and analyze exhaustion markers expression and cytokine production on CAR T cells of different patients. It could also help to coculture the CAR T cells and cancer target cells and assess their cytotoxicity using technique like the real time cell analysis. This could help you assess if the persistent CAR T are still functional or exhausted
