hello, we are thinking of starting some ipsc culture and looking through protocols. do you have some ideas or protocols that you can share? thank you!
Sanem
What’s the source of your ipsc Cells? That will help sharing the protocol specifically for the species. We work with Rat/ Mouse ipscs.
we have plans to work with human endothelial ipscs

In that scenario, you can refer to these Published Protocols:
Patsch et al., 2015 — “Generation of vascular endothelial and smooth muscle cells from human pluripotent stem cells”
Cell Stem Cell
DOI: 10.1016/j.stem.2014.12.001
This is one of the foundational protocols for deriving ECs from iPSCs using small molecules.
Lian et al., 2014 — “Efficient differentiation of human pluripotent stem cells to endothelial progenitors via small-molecule activation of WNT signaling”
PNAS
DOI: 10.1073/pnas.1400294111
Orlova et al., 2014 — “Functionality of endothelial cells and pericytes from human pluripotent stem cells in 3D microvessels”
Stem Cell Reports
DOI: 10.1016/j.stemcr.2014.09.010
Hope these will help you going forward. Let us know if you need further help.
Keep Posting!!

In addition to protocols, you will be required to establish a number of markers which are for Endothelial Cells and for Pluripotency. Your cells should be tested for both presence and absence of following markers: Positive: CD31, CD144, VEGFR2 (KDR), CD34, vWF, eNOS
Negative: OCT4, NANOG, SOX2
Good Luck ahead!
