How do I improve the signal-to-noise ratio (SNR) in my fluorescence microscopy images without increasing photobleaching?

There are multiple ways to achieve this.
- Brighter fluorophores: Choose dyes with high quantum yield and photostability (e.g., Alexa Fluor, Atto dyes).
- Use antifade reagents: Mounting media like ProLong Gold or VECTASHIELD reduce photobleaching and stabilize fluorescence.
- Lower laser power, longer exposure: Instead of blasting fluorophores with high intensity, reduce illumination and increase camera exposure time. This improves signal without excess bleaching.
- Use higher numerical aperture (NA) objectives: Higher NA collects more light, improving signal capture efficiency.
- Proper sample washing: Minimizes unbound fluorophores that contribute to background.
