I’m facing a problem here where I run HPLC using the same method as previously run by my colleague in the same laboratory. But, my peak areas were decreased as compared to hers. We are using Fluorescence detector. We had troubleshoot by checking the autosampler and also changing to the new xenon lamp (as the previous one almost reached to maximum usage of 2000hrs).
I tried to do injection by using autosampler and manual injection and make comparison. But the results are still the same.
After changing to the new lamp, still the peak areas are low.
Before I change to new column, may I have suggestion from expertist about this problem?
