Getting rid of RBCs

Hello

I’m trying to make primary culture of larynx tumour tissue. But unfortunately I can’t remove erythrocytes on my flasks. Do you have any idea how to get rid of them?

You can get rid of erythrocytes by mutiple ways

  1. Give extra washes with PBS prior to extraction of cells
  2. Use a RBC Lysis solution - which usually is a mixture of Ammonium Chloride (155mM), EDTA (0.1mM) and Sodium bicarbonate (10mM)
  3. Perform a density gradient centrifugation when you are isolating cells by using Ficoll or Picoll. You’ll get two separate layers with RBC and cells from laryngeal tissue.
